Appreciation is increasing for the relevance of tumor cell-extrinsic mechanisms to drug resistance observed in a wide range of cancer types. These mechanisms can involve complex cell-cell interactions in tissue microenvironments, raising new challenges with respect to elucidation and understanding – but at the same time also raising new opportunities for address using therapeutic combinations aimed at diverse cell targets. In this seminar, I will present efforts by our laboratory in partnership with various collaborators focused on these challenges and opportunities, demonstrating value of multi-variate systems analysis approaches that closely integrate experiment and computational modeling.
